Let me ask you this: Did you store your bike with the battery in the bike over winter? When you say it wants to start but doesn't, do you mean the engine is turning over but not firing, or not even turning over?

RE: bike wont start
I'd say you need to recharge your battery for starters.
If you are twisting the throttle while trying to start it, the engine is likely flooded, causing a very hard start. Try disengaging the choke fully, and just crank the engine over about 6-8 seconds at a time without using any throttle. I have found that the 250R is very easy to flood. I've done it a few times myself without ever realizing I was doing it.
